    Bacterial heparinase III, also known as heparin lyase III, is a enzyme cloned from Flavobacterium heparinum and expressed in E. coli. Heparinase III can degrade heparan sulfate and serves as a powerful tool for the study of heparan sulfate and for quality control of heparin preparations. This reaction yields oligosaccharide products containing unsaturated uronic acids, which can be detected by UV spectroscopy at 232 nm.

    Heparinase III cleaves the glycosidic bonds between an amino sugar and an iduronic acid or glucuronic acid residue, yielding primarily disaccharides. This enzyme is inactive toward heparin and low-molecular-weight heparins.

    Product Applications

    In pharmaceutical quality control, heparinase III can be used for the 1,6‑ring‑closure assay of enoxaparin sodium and for disaccharide profiling of heparin sodium; when combined with heparinase I and heparinase II, it is employed in the preparation of low‑molecular‑weight heparin (LMWH).
